Aquatic ecosystems support high biodiversity but are difficult to survey with conventional methods. Environmental DNA (eDNA) has emerged as a powerful, non¿invasive tool for monitoring aquatic biodiversity. This Reprint collects original research and case studies on using eDNA to map and monitor biodiversity in hotspot habitats such as rivers, lakes, reservoirs, estuaries and protected areas. Topics include workflow optimisation, primer and database evaluation, comparisons with traditional surveys, biomass estimation, and surveillance of invasive and endangered species. Together, these studies show how eDNA is transforming aquatic ecology and offer a practical reference for researchers and conservation managers.
